Failure to deactivate the default mode network indicates a possible endophenotype of autism

چکیده
UNLABELLED BACKGROUND Reduced activity during cognitively demanding tasks has been reported in the default mode network in typically developing controls and individuals with autism. However, no study has investigated the default mode network (DMN) in first-degree relatives of those with autism (such as siblings) and it is not known whether atypical activation of the DMN is specific to autism or whether it is also present in unaffected relatives. Here we use functional magnetic resonance imaging to investigate the pattern of task-related deactivation during completion of a visual search task, the Embedded Figures Task, in teenagers with autism, their unaffected siblings and typically developing controls. FINDINGS We identified striking reductions in deactivation during the Embedded Figures Task in unaffected siblings compared to controls in brain regions corresponding to the default mode network. Adolescents with autism and their unaffected siblings similarly failed to deactivate regions, including posterior cingulate and bilateral inferior parietal cortex. CONCLUSIONS This suggests that a failure to deactivate these regions is a functional endophenotype of autism, related to familial risk for the condition shared between individuals with autism and their siblings.

Default mode network segregation and social deficits in autism spectrum disorder: Evidence from non-medicated children
Functional pathology of the default mode network is posited to be central to social-cognitive impairment in autism spectrum disorders (ASD). Altered functional connectivity of the default mode network's midline core may be a potential endophenotype for social deficits in ASD. Generalizability from prior studies is limited by inclusion of medicated participants and by methods favoring restricted...
متن کاملFailure to deactivate in autism: the co-constitution of self and other.
A new brain imaging study demonstrates that patients with autism have a strikingly different pattern of brain activity compared with control subjects. During cognitive tasks, cortical areas known as the "default state" network--areas that have been implicated in both self-referential processing and processing of socially relevant information--typically reduce their brain activity. In patients w...
متن کاملWhole-brain functional hypoconnectivity as an endophenotype of autism in adolescents
Endophenotypes are heritable and quantifiable markers that may assist in the identification of the complex genetic underpinnings of psychiatric conditions. Here we examined global hypoconnectivity as an endophenotype of autism spectrum conditions (ASCs). We studied well-matched groups of adolescent males with autism, genetically-related siblings of individuals with autism, and typically-develop...

PET imaging of Alzheimer's disease (AD) patients demonstrates Abeta-protein accumulation in a default mode network. In this issue of Neuron, Sperling and colleagues show that cognitively intact individuals with Abeta in this network fail to deactivate it during memory tasks, a finding with important implications for cognitive aging and early detection of AD.

Autism spectrum disorder refers to a neurodevelopmental condition primarily characterized by deficits in social cognition and behavior. Subclinically, autistic features are supposed to be present in healthy humans and can be quantified using the Autism Quotient (AQ). Here, we investigated a potential relationship between AQ and neural correlates of social and monetary reward processing, using f...
